I am worry about the mother have not pee and poo almost 24hrs after a singleton born,she drink and eat normal,she don't to leave the nest one second,pl advise.

She will eventually. I'd be more concerned if the puppy weren't peeing and pooping!
Take momma outside for a nice walk, and keep her out there until something happens! It's likely her concern for her pup that's preventing her from going.
Is the mother eating and drinking at all?

Yes, she is drinking and eating normal.

That's good to know. Well, what goes in, must come out...eventually! 
I'm glad to hear that her mothering instincts are so strong that she doesn't want to leave her baby. But take her outside regularly, and eventually she will have to go potty.
