Is it safe to do an ultra sound on a pregnant female and when is the best possible time?

Yes, its safe.
Is possible to see the eggs early at 21 days, but 25 days is more clear. At 32 days the heartbeat can be seen.
If the whelping is delayed is very usefull to check for puppies health.

i ultra sound on the 50 day just to see how many puppys........my vet says 50 day is the best...

28 days I have found to be best as further on in pregnancy puppies hide behind each other!

Sure it¿s save, and as Arko already wrote around the 25th day you can see if the female is pregnant.
And I can tell you, it has become very usual in Germany that you don¿t get the "Deckschein" (sorry I don¿t know the english word) from the male-owner until you have conformation of the pregnancy of your female by a vet by doing ultra sound.

I do ultrasound with qualified vet at 30 days give or take a few - he gets an accurate count at that time, and can check all heartbeats. He is always within 1 puppy in his count - the farther along in pregnancy, the harder to get a count because puppies get too big and can hide behind each other, as someone also said.

I also use ultrasounds at the 25-30day mark. Just like christmas I can never wait to open the present. not only does it give you a half way decent count on puppies it helps me gear up to get ready for the litter. After knowing she is positively pregnant, I start getting her acustom to the welping box and get ready for the birth. It also helps me clear my schedule around that time as I am always present for the births.
