My female had progestrone level of 1.77ng/ml on 8th day & she had successful matimg, on 10th day (yesterday) her levels was 4.08ng/ml & had mating again. Now should i go for another set of test or go directly for mating today or tomorrow???

Tomorrow , today is covered from yesterday, semen lives minimum 3-4 days inside her, plus give male a chance to strengthen sperm production...good luck

Here are the numbers:
0-2 nmol/L Baseline concentrations too early to estimate ovulation
3 - 6 nmol /L Minimum 2 days before ovulation expected however result of 3 - 4 nmol/L may persist for a week or longer before increasing. Earliiest estimating breeding 4 - 6 days but could be longer (re test needed)
7 - 12 nmol/L Minimum 1 day before ovulation. Estimated window for breeding 3 - 5 days but could be longer ( re test recommended)
13 - 18 nmol/L Ovulation impending or just occurred. Estimated breeding window 2 - 4 days 
19 - 31 nmol/L Ovulation recently occurred. Estimated breeding window 1 - 3 days
32 - 64 nmol/L Ova have matured, optimal potential for fertility. Estimated breeding window 0 - 2 days
65 - 90 nmol/L Ova have matured but aging, decreased potential for fertility. Breed at once
Above 90 nmol/L Too late

Note the numbers in the post by Miro are given in nmol/L whereas the original post is ng/mL.   They're not the same scale.

General rule of thumb is that the prime time to breed is about 48 hours after progesterone hits 5 ng/ml. You can breed before then because the semen is viable for up to 4 days in the female. You have a short window after that period--say 2 more days.
  • The initial progesterone rise during estrus in the bitch coincides with the LH peak. 
  • Before the LH peak the progesterone is < 1.0 ng/ml. 
  • On the day of the LH peak, progesterone rises to around 1.5 - 2.0 ng/ml. Thereafter the progesterone is > 5 ng/ml. 
  • Using our lab ovulation occurs at about 3.8-5.0 ng/ml (11.7 nmol/L) of progesterone (including the day of ovulation and the 24 hours it takes to ovulate) and the best days to breed occur at about 17.6-24.1 ng/ml (54-74 nmol/L) of progesterone.

On 10th day her levels were 4.08ng /ml and vet said there is no need for getting more progesterone test, so on 12th day we mated her again without getting progesterone.
Is this ok?

All vets agree that the best days for natural mating or AI with fresh or chilled semen is day 1 and 3 (or day 2 and 4) after ovulation. Ovulation starts two days after LH surge when P approaches 2.0 ng/ml.  

But there are two schools of thoughts.

1. LH surge most reliable  therefore about P=2.0ng/ml is LH surge

2. Ovulation is P=5mg/ml.

The practical difference between the two thoughts is likely 12 to 24 hours.

Why did you allow mating so early? Your first mating was day of LH surge. Far too early from ideal, but report is that a single natural mating any time between day of LH surge and 5 days after LH surge results in the same conception rate and litter size.

You do not need to do any more P test, but you should count the days correctly.

In your case, third and fifth day (or 4th or 6th day) after P=1.77 would have been the best two mating days. 

If you trust ovulation more, then 1 and 3 days (or 2 and 4 days) after P=4 would have been the best two mating days.

Thanks HighDesert GSD, well last time i remember when she had her first (12thday) mating, her progesterone level was 12ng/ml, so i thought her ideal days might comes early. So i decided to start getting progest. from 8th day and mating too, thats why i get early. Her last mating was on 12th day (toatl 3 matings 8th, 10th and 12th day). Keeping in mind that sperms last for approx 3-6 viable in natural mating thats why i didnt go for further mating.
