Views on what the dog is saying. Is it serious? Is it prey/defence? I would like to get educated views.

I agree there is something unsure going on. I am going to guess, it was you or someone familiar behind the camera, but the dog wasn't too sure if the camera was a threat or not, so that created confusion. The fact that the dog was confined would/could add to the stress. I would not put my hand through the window, that is for sure. The tail movement suggested something was familiar to the dog, I think. The forward movement with the show of teeth, intensity of the eyes was definitely a challenge. IMHO

He looks a little perplexed, but like Beetree said, I wouldn't stick my hand in that car! Just because a dog may not be familar and conditioned to a certain situation, and may appear a little unsure, does NOT mean the dog won't bite. I respectfully disagree that it's a play bark.

It will be interesting to see what everyone has to say about this; I think he's throwing a tantrum at someone or something going by or too close tot he truck. I would not get near the truck personally or stick my hand in to pet him. Learned that years ago; a male Samoyed almost got me when I reached in to pet him, he actually knew me too and almost bit me. Ugh... This dog has that similar look.
Maybe someone gave him the guard command, did the post man walk by; or the UPS guy stop?? Gee's... Nan

I don't see it as an aggressive bark. He is not barking loud and at one point whines in the middle of barking.
He wants something. Possible to go with his master or he wants to go play, maybe he sees other dogs.
Also there is a series of two barks together. All of the GSDs I've had barked twice (or a series of two barks together) when they wanted something.

The pitch of the bark was high. At one point there was very brief whinning. If the dog was guarding the car and barking at a stranger it should have been a much deeper pitch. The tail was wagging but, there was a slight growl in the bark.
